Assay requires 100-200 ng per array depending on ploidy; please send at least 200 ng at 10 ng/µL or above.
The assay requires 100 ng (human), 150 ng (diploid animals and plants) or 200 ng (polyploid animals and plants) per array. In the lab each sample is diluted to 20 µL at 5, 7.5 or 10 ng/µL respectively (Thermo Fisher, MAN0017720).
Please send at least 200 ng per sample at 10 ng/µL or above, in at least 20 µL. We recommend 500 ng where available, so a repeat run is possible without resubmission.
Dissolved in nuclease-free water or reduced-EDTA TE buffer (10 mM Tris-HCl pH 8.0, 0.1 mM EDTA). Standard TE with 1 mM EDTA is an inhibitor and not suitable.
Concentration determined fluorometrically (Qubit or PicoGreen). DNA must be double-stranded, so no boiling or strong denaturants.
Not degraded. Free of polymerase inhibitors, in particular heme from blood, high salt and chelating agents. OD260/280 between 1.8 and 2.0, OD260/230 above 1.5; please clean up samples below these values before shipping.
No material from FFPE blocks.
